In vitro Propagation of Pinus roxburghii Sargent using Embryonic Material

Abstract

Attempts were made to induce adventitious buds on whole embryo and excised cotyledons from partially germinated seeds of Pinus roxburghii Sargent. The whole embryo and excised cotyledons were established on solid MS medium supplemented with 4.0 and 3.0 mg/1 BAP, respectively. The embryos showed elongation and exhibited meristimatic activity after 12 weeks of establishment.

The excised cotyledons exhibited exhibited meristimatic activity after 4 weeks of incubation on solid MS medium supplemented with 3.0 mg/1 BAP and 0.5 mg/1 IBA. The adventitious buds initiated on the surface of cotyledons after 12 weeks on the medium which showed swelling and elongation.
